You'll need to provide a much better picture than that for help. Get the mark in focus, maybe with something white in the background.

This is a detail of the spot...what is it? In your opinion visible in a shot? (For the moment I have not the adapter)

It shouldn't be visible in normal conditions, perhaps when stopped down to f/22 it might appear.
But I would check if it's fungus, and in that case definitely return the lens to the buyer.
The price was ok for an AE, but not a super bargain, so if the lens is not of your full satisfaction, I'd say you
should consider returning the lens then add some further 100 to the sum to get a MM copy.
But then that's what I would probably do, but your mind could be different.

Looks more like a spec of paint off the inside of the lens barrel than fungus. Should be easy to remove if you can strip the lens down.
